What is Amp^R?
A gene that codes for Ampicillin resistance
What is Pbad?
Can be turned on+off under certain conditions, GFP promoter
What is the Ori?
Origin of replication
What is the GFP gene?
Codes for protein that glows florescent green (Green Florescent Protein)
What is araC?
Acts as a regulator for the GFP gene, switch
What is the purpose of the plate with no additives?
Acts as a negative control. No growth here=mislabeled plate? dead cells?
What is a competent bacteria?
bacteria that have the ability to take up foreign DNA from their environment through a process called transformation.

What's 1?
araC

Whats 2?
Pbad

Whats 3?
GFP

Whats 4?
Amp^r

Whats 5?
Ori (origin)
Why can bacteria with pGLO grow in Ampicillin plates?
pGLO containing amp^r allows the bacteria to resist dying, while those without pGLO will die.
What does warming the bacteria with pGLO plasmid DNA do? >42 degrees C
Allows competent to open, letting in the pGLO DNA
What does cooling the bacteria with pGLO plasmid DNA do? <42 degrees C
Allows the bacteria to close, there for trapping the pGLO DNA
