What is the aim of this experiment?
To isolate DNA from a plant tissue
What are the materials required?
Onom
Washing up liquid
Blender
Distilled water
Sharp knife
Table salt
Protease enzyme trypsin
Coffee filter paper
Ethanol at freezer temp
Droppers
Water bath
Spatula
Ice water bath
Plastic syringe
2 beakers
Radiated
Boiling tube and test tube rack
Graduated cylinder
Retort stand
Glass rod
Balance
Funnel
How do you start the experiment
Add 3g table salt to 10cm cubed of washing up liquid in a beaker and fill up to 100cm cubed with distiller water
What do you add to the salty washing up solution?
Chopped up onion
Where should you place the beaker after adding in the onion?
After bath at 60 degrees for exactly 15 minutes
How do you cool the mixture after the water bath?
Stand the beaker in an ice bath for 5 minutes stirring frequently
How long should you blend the cooled mixture for?
No longer than 3 seconds
How much filtrate should be added to the boiling tube?
10cm cubed
What do you add to the filtrate in the boiling tube?
2-3 drops of the enzyme protease
After you add the protease, how should you add the next chemical to the filtrate?
Trickle 10cm cubed of ethanol (freezer temp) down the side of the boiling tube - forms a layer on top of the filtrate
What do you do with the boiling tube after all chemicals are added?
Leave without disturbing for a few minutes
How do you draw DNA from the test tube
Using a glass rod
