Identifies myeloid cells with lipid granules.

Myeloid cells will stain strong and Monos will stain weakly.

Specific esterase (CAE)

Helps differentiate granulocytes from monocytes.

Granulocytes will stain strongly

Non-specific esterase (NSE)

Used to help identify monocytes and their precursors

Monocytes and monoblastic stain strong positive

Helps distinguish AML M4, M5a and M5b

Acid Phosphatase

Helps diagnose T-cell leukemia (T-ALL)

T-cells stains strongly

Monocytes stain weakly

TRAP

Helps stain Hairy Cell Leukemia (HCL)

Stains bright red

Periodic Acid Schiff (PAS)

Detects glycogen and neutral mucins.

Lymphs and Erythroblasts stain strongly

Helps diagnose AML-M6 and slightly AML-M7

Detects early lymphoid lineage and helps diagnose ALL (B and T cell)

Lymphoblasts stain strongly
