Step 1
Assemble equipment and materials.

Wash hands or use alcohol-based hand-rub for hand hygiene.

Obtain recently collected urine specimen.

Complete demographic information on the Laboratory Form.

Mix urine by gently swirling and pour about 10 mL into a clear conical centrifuge tube.

Observe the color of the urine and record it on the Laboratory Report Form.

Observe and record the transparency of the urine.

Note the odor of the urine and record it in the comment section.
Step 10a
Place one drop of distilled water on the refractometer glass plate and close gently.
Step 10b
Look through the ocular and read the specific gravity.
Step 10c
If water does not read 1.000, calibrate the refractometer.
Step 10d
Disinfect the refractometer by rinsing with 1-2 drops of water and drying with lab tissue.
Step 10e
Place one drop of well-mixed urine on the refractometer glass plate using a pipette.
Step 10f
Close the lid gently.
Step 10g
Look through the ocular and read the specific gravity on the scale.
Step 11
Record the specific gravity accurately on the Laboratory Report Form.
Step 12
Clean the refractometer with lens paper and disinfectant, then return it to storage.
Step 13
Verbalize disposing of the urine specimen.
Step 14
Clean the work area with disinfectant.
Step 15
Dispose of PPE appropriately.
Step 16
