Put a drop of iodine into every well of a spotting tile
Place the Bunsen burner on a heat proof mat, and a tripod and gauze over the Bunsen burner. Heat a beaker of water until its 35 degrees. Try to keep the temperature constant throughout the experiment
Use a syringe to add 1cm3 of amylase solution and 1cm3 of a buffer solution with a pH of 5 to the boiling tube. Use test tube holders. Put the test tube in a beaker of water and wait for 5 minutes.
Next use a different syringe and add 5cm3 of a starch solution to be boiling tube.
Immediately mix the contents of the boiling tube and start a stopwatch
Use continuous sampling to record how long it takes for amylase to break down all the starch. To do this take a fresh sample from the boiling tube every 30 seconds and put a drop into the well. When the iodine solution no longer changes colour no starch is present.
Repeat the experiment with buffer solutions with different pH levels to see how pH affects the time taken for starch to break down
