acid

Acid Fast Stains – Kinyoun Method
Create a heat fixed smear
• Cover the smear with Kinyoun carbolfuchsin for 5-10
minutes
• Rinse with distilled water
• Decolorize with acid-alcohol until the run-off is clear
• Rinse with distilled water
• Apply methylene blue counterstain for 1 minute
• Rinse with distilled water.
• Blot on Bibulous paper
• Observe under oil immersion
