Buffer P2

NaOH + SDS, lyses cellular membrane and denature proteins

Buffer EB

Low-salt buffer that elutes DNA from the silica membrane

Buffer PE

Wash buffer 2: removes ions and other impurities from the plasmid DNA. Contains ethanol and salts, to ensure the plasmid DNA stays attached to the column.

Buffer N3

High-salt buffer that precipitates bacterial cell contents (chromosomal DNA, proteins, membrane, etc..)

Buffer P1

Tris, EDTA, RNAse, Destabilizes outer membrane of bacterial cells

Buffer PB

Wash buffer 1; removes DNases which may degrade the plasmid DNA. Contains isopropanol and salts, which ensure that the plasmid DNA stays attached to the column.
