What is the equipment needed to experiment the effect of pH on amylase?
Spotting tile
Pipette
3 test tubes
2cm cubed of amylase solution, starch solution & buffer solution
Iodine
Stopwatch
What is the method for testing the effect of pH on amylase?
Place one drop of iodine in each spot in the spotting tile
Mix all the testubes together and start a stopwatch
Every 30 seconds, take a drop from the mixture and observe
Eventually, the iodine will stop turning blue/black as there won’t be starch present anymore. Record how long this took.
Repeat the experiment multiple times using different pHs of buffer solution.
What happens to amylase at pH1?
The active site denatures, meaning it can no longer bind to starch molecules.
What are 2 problems with this experiment?
Taking samples every 30 seconds only gives an approximate time. It would be better to take them every 10.
It isn’t always obvious when iodine stops changing colour. It would be better to ask multiple people to observe.
