Acetone

• Used at cold temperatures (-5-4 °C)

• Fixative used when studying water soluble enzymes (Phosphatases and Lipases)

• Fixative used on brain for rabies studies

Trichloroacetic Acid

• Both a fixative and weak decalcifying agent

• Can soften dense fibrous tissue

Heat Fixation

• Thermal coagulation of proteins

• Frozen Section and Bacteriologic Smears

• fresh, unfixed that will undergo frozen & immunofluorescence RT 5 days

• NOT SUITABLE for flow CYTO Or FISH

Microwave Edition

• issue heated in a very short time allowing cellular process that occur rapidly to be studies

• Disadvantage: Can only penetrate tissues 10-15mm thick

• No cross-linking of proteins

• Spores and Pathogens may remain viable in tissues
