Step 1
Label 8 bottles A-H
Step 2
Transfer 10ml of stock solution into bottle A. Pour 9ml of water to bottles B-H
Step 3
Drop 1ml of IAA from A to B and stir. Repeat this trasnferring 1ml from B to C, C to D etc until G
Step 4
Remove 1ml from bottle G and dispose of it
Step 5
Place acetate grids in the lid of 8 petri dishes
Step 6
Place cress seeds along the grid lines of each
Step 7
Place filter paper on the seeds in each dish
Step 8
Add a few drops of each IAA solution to each filter paper.
Step 9
Cover filter papers with wool
Step 10
Pour rest of IAA solutions onto each piece of cotton wool
Step 11
Tape the petri dishes shut
Step 12
Stand petri dishes horizontally
Step 13
Leave dishes in an incubator at 20-25 degrees for a few days
Step 14
Use acetate grid to measure shoots and roots and record results
