Tolerance
A decrease in drug effect with repeated administration or an increase in the dose required to produce a similar effect.
Types of tolerance
acute, pharamcokinetic, pharmacodynamic, functional disturbance, behavioural
Pharmacokinetic tolerance
occurs when the body becomes more efficient at metabolizing a drug
Acute tolerance
develops rapidly, often within a single drug administration
Pharmacodynamic tolerance
involves changes at the site of drug action, changes in receptor sensibility: upregulation, downregulation
Functional disturbance tolerance
changes in physiological functions that counter the drug’s effect
Behavioural tolerance
based on drug discrimination learning
