Structural stains

Can be used to identify and study the structure of batceria

Endospores

  • Known as resting bodies because they don’t metabolize and are resistant to heating, various chemicals, and harsh environmental conditions

  • Formed when water or essential nutrients are not available

  • Can remain dormant, but can also return to its vegetative or growing state

  • Impermeable to most stains, so heat is usually applied, also meaning that the stain cannot be washed out (retains malachite green)

  • In other words, endospores are highly resistant, dormant structures produced by certain bacteria that can survive harsh conditions

Endospore Stain Reagents

  • Malachite green (Primary stain)

  • Safranin (Counterstain)

Malachite green stains the endospore green, while Safranin stains the vegetative cells pink or red

Endospore Stain Procedure

To differentiate between endospores and vegetative cells within bacteria
