Structural stains
Can be used to identify and study the structure of batceria
Endospores
Known as resting bodies because they don’t metabolize and are resistant to heating, various chemicals, and harsh environmental conditions
Formed when water or essential nutrients are not available
Can remain dormant, but can also return to its vegetative or growing state
Impermeable to most stains, so heat is usually applied, also meaning that the stain cannot be washed out (retains malachite green)
In other words, endospores are highly resistant, dormant structures produced by certain bacteria that can survive harsh conditions
Endospore Stain Reagents
Malachite green (Primary stain)
Safranin (Counterstain)
Malachite green stains the endospore green, while Safranin stains the vegetative cells pink or red
Endospore Stain Procedure
To differentiate between endospores and vegetative cells within bacteria
