What is resolution?
ability to completely separate two objects in the microscopic field
What is parfocal?
ability to change from low power to high power objectives with only fine focus adjustment

How does wavelength affect resolution?
shorter wavelength = greater resolution
what color filters does a microscope use? why?
Blue filter
*has the lowest wavelength of all visible light
-creates the best resolution

Why does oil help increase resolution?
Since oil has same refractory index as glass, it eliminates the refraction index
*allows for very high magnification (1000x) and still good resolution
What are the 5 broad steps of finding things in the microscope?
1. clean off the lenses
2. find a reference point
3. start on low power (10x)
4. switch to high power (40x)
5. switch to oil power (100x)
How can one find a reference point?
looks for clumps or the edge of the cover slip
*can also draw a circle on the slide
What should be done with finding things on low power?
Start with the stage all the way done and slowly move up
*move the stage to make sure sample is in focus (and not just part of the microscope)
What should be done at high dry power?
Only use the fine adjustment knob
*change the condenser to the objective (40x)
What can be done on 100x power?
Add oil and only use the fine adjustment
*change the condenser
