1. Cut DNA: Restriction enzymes cut open the bacterial plasmid vector and isolate/cut the human insulin gene.
2. Join Plasmid and insulin gene: DNA ligase seals the human gene into the plasmid, forming a recombinant plasmid (ligation).
3. Deliver: The recombinant plasmid is introduced into a host bacterium like E. coli through transformation.
4. Select: Antibiotic resistance markers or screening methods are used to identify and select only the bacteria that successfully took up the plasmid.
5. Grow: The selected recombinant bacteria are cultured and fermented on a large scale to multiply and produce the insulin protein
