Insert/Chromosomal DNA
DNA fragment of interest for cloning. Obtained from PCR
Cloning Vector/Vector DNA
Vehicle that transports foreign material into DNA. Normally plasmids. Contains an origin of replication (site of DNA replication), MCS (unique sites for restriction enzymes to cut), and markers (identifies cells with the vector)
Restriction Enzymes
Proteins that cut DNA at specific sequences. They are essential for creating recombinant DNA by processing the insertion of foreign DNA into cloning vectors. Forms sticky and blunt ends by cutting target gene and vector.
DNA Ligase
The molecular glue that brings together the target gene and vector by catalyzing formation of phosphodiester bonds
