My name is Ngà. I am currently working in the Microbiology – Raw Materials team of the QC Department at B. Braun Vietnam.

Today, I would like to introduce the bacterial endotoxin testing method using the gel clot technique.

This method is used to determine the level of bacterial endotoxins in a product based on the clotting reaction between endotoxins and the lysate reagent.

First, we prepare the equipment, reagents, and test samples. All items must be within their expiration date.

Next, we prepare the control samples, including the Negative Control (NC), the Positive Control using standard endotoxin solution (PC), and the Positive Product Control (PPC).

After that, we proceed with the test:

• Treat the samples according to section 6.2.2 of SOP 019

• Mix an equal volume of 100 microliters of NC, PC, PPC, and the diluted test sample with 100 microliters of lysate reagent

• Prepare two tubes for each sample and incubate at 37 ± 1°C for 60 ± 2 minutes

After incubation, read the results and draw conclusions:

The test is considered valid when the NC shows no clot formation (negative), while the PC and PPC show clot formation (positive).

Hôm nay tôi xin giới thiệu về phương pháp xác định nội độc tố vi khuẩn bằng phương pháp tạo gel.

 Đây là phương pháp xác định hàm lượng nội độc tố vi khuẩn trong sản phẩm dựa trên sự đông tụ của nội độc tố và thuốc thử lysate.

Trước tiên chúng ta chuẩn bị thiết bị, thuốc thử và mẫu cần kiểm tra. Tất cả phải còn hạn sử dụng.

Tiếp theo chúng ta chuẩn bị mẫu đối chứng ( NC), dung dịch nội độc tố chuẩn gốc ( PC), mẫu kiểm tra sự dương tính trong sản phẩm ( PPC).

Sau đó ta tiến hành thử nghiệm:

+ Xử lý mẫu theo mục 6.2.2 của SOP 019

+ Trộn đồng thể tích 100 microlit NC, PC, PPC và mẫu thử đã pha loãng với 100 microlit thuốc thủ lysate

+ Lặp lại 2 ống cho mỗi loại, ủ ở 37 độ c cộng trừ 1 độ,  trong thời gian 60 phút cộng trừ 2 phút.

Khi quá trình ủ kết thúc đọc kết quả và đưa ra kết luận:

Phép thử đạt yêu cầu khi NC, mẫu đều dương tính và PC, PCC đều âm tính
