Albumin Solubility in Distilled Water
The behavior of albumin as a water-soluble globular protein that remains fully dissolved without any change in turbidity when diluted with distilled water.
Alkali-Albuminates
Highly soluble protein complexes formed when strong bases like NaOH denature albumin and induce strong negative charges on the molecules, causing them to repel each other and remain clear or slightly translucent.
Isoelectric Point Disruption by Acid
The process where strong mineral acids like HCl drastically lower pH below a protein's isoelectric point, disrupting salt bridges and hydrogen bonds to cause unfolding and coagulation into a visible white precipitate.
Heat Denaturation of Albumin
The disruption of weak bonds (hydrogen, ionic, hydrophobic) caused by high temperatures increasing kinetic energy, destroying secondary and tertiary protein structures while leaving the primary amino acid sequence intact.
Coagulation (Gelation)
The physical phase during protein denaturation where unfolded protein chains aggregate to conceal exposed hydrophobic interior regions, trapping water and forming an insoluble solid or semi-solid network.
Heat Coagulation Test
A standard laboratory test for proteins where heating top portions of a sample causes turbidity, and 1% acetic acid is added to dissolve phosphoprotein if present.
Organic Solvent Denaturation
Protein precipitation induced by agents like 95% ethyl alcohol, which interfere with internal hydrophobic interactions and form new intermolecular hydrogen bonds that collapse the tertiary structure.
Concentrated Nitric Acid Effect
Irreversible denaturation and precipitation of albumin caused by extreme pH changes and high hydrogen ion concentration, forming a white precipitate that does not redissolve upon adding excess acid.
Alkaloidal Reagents
Anionic precipitants carrying strong negative charges that neutralize positively charged amino groups of proteins in an acidic medium, forcing irreversible denaturation and precipitation.
Tannic Acid Reaction
An alkaloidal reagent test that produces a heavy, murky yellowish-brown to dark-brown protein-tannate salt complex with high precipitation efficiency.
Picric Acid Reaction
An alkaloidal reagent test based on Esbach's reagent principle that combines with protein to form a bright yellow to tangerine-colored insoluble protein-picrate precipitate.
Trichloroacetic Acid (TCA)
A powerful dehydrating and neutralizing alkaloidal reagent (10%) that completely unfolds albumin to immediately form a dense, opaque milky-white protein-trichloroacetate salt precipitate.
Action of Heavy Metal Ions
Irreversible protein denaturation and coagulation caused by metal cations such as Ag+ from 5%AgNO3, resulting in the immediate formation of a dense white precipitate.
Pepsin
A digestive enzyme that catalyzes the enzymatic digestion of proteins like egg white when combined with dilute HCl (0.25%) at an optimal temperature of 37∘C to 38∘C.
