What are the components of xylose lysine deoxycholate (XLD) media?
Lysine, xylose, lactose, sucrose, ferric ammonium citrate.
Sodium deoxycholate inhibits Gram positive organisms; phenol red as indicator

India ink procedure
Place a small drop of specimen (ex. CSF, culture) onto a clean glass slide

Why do Gram positive bacterial cells stain purple?
The thick peptidoglycan cell wall traps the crystal violet stain and resists decolorization under alcohol.

The large lipid content in the cell walls is removed by the alcohol wash along with the CV stain.
