LB Agar
Non-selective, non-differential
Grows E.coli for molecular biology applications

Trypticase Soy Agar (TSA or TSAYE)
Non-selective, non-differential
Serves as base for enriched medias

Blood Agar (BAP)
Non-selective, differential, enriched
Grows fastidious microorganisms
Differential: Detects hemolytic activity
Differentiates Streptococcus strains

Hemolysis
Breakdown of RBC
Alpha hemolysis
Partial lysis of RBC; greenish-brown color colonies

Beta hemolysis
Complete lysis of RBC; distinct zone of clearing around colony

Gamma hemolysis
No lysis

Chocolate Agar (CHOC or CBA)
Non-selective, non-differential, enriched
Derivative of blood agar; made of already LYSED RBC (by heat)
Enriched: fastidious and pathogenic bacteria

Colombia CNA Agar
Selective, differential, enriched
Selective: antimicrobial agents (colistin and nalidixic acid); FOR gram POSITIVE
Differential: blood —> hemolytic patterns

Mannitol Salt Agar (MSA)
Selective, differential
Selective: NaCl (7.5-10%); selective FOR gram POSITIVE
Differential: Mannitol & pH (phenol red); differentiates pathogenic strains of Staph
Pathogenic FERMENTS mannitol → agar Δ color from red (>8.2) to yellow (<6.8)

MacConkey Agar
Selective, differential
Selective: Crystal violet & bile salts RESTRICT gram POSITIVE
Differential: Lactose & pH; red means acidic, which means lactose is fermented
Colonies change color AND base (surrounding area) sometimes
pH > 8.0 → agar turns yellow; pH <6.8 → agar turns darker red or pink
Pale red

Sorbitol-MacConkey Agar (SMAC)
Selective, differential
Differential: detects pathogenic strain of E. coli (O157:H7)
E. coli O157:H7 unable to ferment sorbitol → colorless/white colonies instead of red
Contains sorbitol instead of lactose

Eosin Methylene Blue Agar (EMB)
Selective, differential
Selective: Eosin & methylene blue RESTRICT gram POSTIVE
Differential: Lactose;
Non-ferm → white/tan colonies; lactose-ferm → dark (purple to black) colonies
E. coli has metallic green sheen (a LOT of acid)

Agar
Polysaccharide made from algae/seaweed
Microbes can’t digest !
How to make agar
Add powdered agar to water (or growth media)
Heat to 100 degrees Celsius so it dissolves (100 degrees retains most of nutrients added to media & won’t damage antibiotics)
Solidify at below 40 degrees Celsius (sensitive ingredients like blood can be added before agar solidifies)
Solid until greater than 100 degrees again !
