What are ligands
Small molecules to proteins, what ever is binds onto the other things
Ligand binding
non-covalent
At binding site, complementary (h-bonds, ion interaction, hydrophobic, van der waals, shape and size, his tag-nickel)
Can be cooperative (sigmoidal, allostery), non-cooperative (hyperbolic)
Ka vs Kd
Association vs dissociation
Kd is just reciprocal of Ka and less ambiguous
Kd
Ligand binding will approach 1
Kd is unit of concentration of ligand needed for half of the protein to be bound
Reflects affinity- reverse relationship (low Kd means strong binding)
