Rules for Microscope Use
Carry with two hands, clean lenses with lens paper only, do not force adjustment knobs, always cover after use, handle cords carefully.
Light Microscope
Uses light to view samples.
Electron Microscope
Uses electron beams for higher magnification.
Eyepiece
Magnifies image from the objective lens, usually 10x.
Nosepiece
Holds and rotates objective lenses.
Objectives
Lenses with varying magnifications: Scanning (4x, red), Low Power (10x, yellow), High Power (40x, blue).
Stage
Platform where the slide is placed.
Coarse Focus Knob
Used for large adjustments when focusing.
Fine Focus Knob
Used for detailed focusing.
Total Magnification
Calculated by multiplying eyepiece magnification by objective lens magnification.
Steps to Use the Microscope
Fixation
Preserves tissues using chemicals for microscopy.
Staining
Involves using dyes like Hematoxylin to stain basophilic structures and Eosin to stain acidophilic structures.
Transmission Electron Microscope (TEM)
Produces a 2D image by passing an electron beam through the specimen.
Scanning Electron Microscope (SEM)
Produces a 3D image by bouncing an electron beam off the specimen surface.
