Glycine (The Achiral Amino Acid)

Why is it achiral? is just a single Hydrogen atom (-H). This means the central alpha-carbon is attached to two identical Hydrogen atoms. Because it does not have four different groups, it loses its chirality.

  • Physical Property: Because it is achiral, it does not have enantiomers (left or right-handed versions), and it does not rotate plane-polarized light (it is optically inactive).

  • Structural Superpower: Being the smallest and least sterically hindered amino acid allows Glycine to fit into incredibly tight spaces in protein structures. It is heavily concentrated in collagen (making up about one-third of its structure) because it allows the triple helix to pack tightly together.

Enantiomers

Because the 19 chiral amino acids have four different groups around the central carbon, they can exist in two different mirror-image forms, called ‘blank’. We designate these as L (Levo/Left) and D (Dextro/Right).

Why do we care about D-amino acids if humans don't use them?

Because antibiotics like Penicillin and Cephalosporins are designed specifically to target and block D-amino acids in the bacterial cell wall. Because human cells strictly use L-amino acids, the antibiotic perfectly kills the bacteria without accidentally attacking human tissue (this is the definition of selective toxicity).

The "Double Chiral" Exceptions

They have the standard chiral alpha-carbon, plus a second chiral carbon located within their R-group side chain.

The Two Double-Chiral Amino Acids:

  1. Isoleucine

  2. Threonine

Double Chiral: memory hack

Examiners love to take a basic rule and test you on the rare exceptions. While 19 amino acids have one chiral center, there are exactly two amino acids that have TWO chiral centers.

Memory Hack: Think of the word IT (Isoleucine, Threonine). IT takes two to tango (two chiral centers). Because they have two chiral centers, these specific amino acids can actually form four different stereoisomers instead of just two.
