Disposal Guidelines
Rules for disposing of laboratory materials including glass, biohazard materials, and standard trash.
Kligler's Iron Agar (KIA)
A medium that differentiates between organisms based on their ability to ferment glucose and lactose, producing a color change.
Ornithine Decarboxylase Test
A test to see if microorganisms can use ornithine as a carbon source, indicated by a color change in the medium.
Xylose Fermentation Test
A test to determine if an organism can ferment xylose, indicated by a pH indicator changing color from red to yellow.
Lysine Decarboxylation Test
A test that assesses the ability of an organism to decarboxylate lysine, indicated by yellow to purple color change.
Bacterial Transformation
The process where a cell takes up and expresses a new piece of genetic material, often resulting in new traits.
Plasmid
A DNA molecule that can replicate independently of chromosomal DNA, often found in bacteria.
Green Fluorescent Protein (GFP)
A protein derived from the Aequorea victoria jellyfish that glows green under UV light.
Antibiotic Selection
The process of using antibiotics in culture media to select for bacteria that have taken up plasmids conferring resistance.
Transformation Solution
A solution that facilitates DNA uptake by neutralizing negative charges on DNA and the cell membrane.
Heat Shock
A step in the transformation process that increases the permeability of the cell membrane to DNA.
Recovery Phase
A period post-transformation where bacteria are allowed to express antibiotic resistance before being plated on selection media.
Objective of the Lab
To transform bacteria with a gene for GFP and observe expression under UV light.
Prelab Assignment
Preparation tasks due before the next lab session, including reporting on chemical germicides.
