pre-compatibility testing

• before issuing compatible bloods, pt must have a valid sample and grouping history

• if not the case, 2 samples will be required to be taken 5 mins apart

• AB screen should also be performed

IAT/full crossmatch

• serological crossmatch is performed using IAT technique

• most common serological crossmatch technique

manual IAT method

• requires 0.8% donor red cell suspension

• 50 uL of red cell suspension and 25 uL of plasma are pipetted into AHG card

• indubated at 37 degrees for 15 mins

• centrifuges

• read results macroscopically

room temp spin XM

• technique is also known as saline spin or immediate spin technique

• used in an emergency to detect ABO incompatibility using manual tube technique

• donor red cell suspension is required in ratio of 2:1 serum to plasma

• solution incubated for 5 mins at room temp

• centrifiged at high speeds

• read macroscopically
