In step 6 of the procedure, you are instructed to place your test the in a water bath and evaporate solvents to 0.2-0.3 mL. Why o you need to concentrate your extract before lacing it on the column?
Concentrating the pigments will increase the yield the extraction
In step 11c of the procedure, you are using hexane t elite b-carotene first and next using acetone to elite chlorophyll. Why?
Because using acetone will result in the elution of the yellow and green pigments w/o separation. These two will move faster through the column due to the solvent being more polar.
Provide analysis of your UV-vis spectra for yellow and green fractions, compare to the spectra found in literature.(Write down -max alums and absorb acne for the two compounds and compare the values with pure compound spectra, search literature)
For acetone our max value was 438nm and absorbable was 0.995
B-carotene peaks at 460 which is close to our value
For hexane max value was 459nm and absorbance was 0170
Chlorophyll peaked t 430nm which is also close to our value
