Osteoporosis

-high risk of fracture
-more than 1.5 million fractures/year
-reduction in bone mass & deterioration of microarchitecture
-associated with accelerated remodeling due to age

Testosterone & Estrogen effects on Bone

-less Testosterone & Estrogen increases bone resorption
-these hormones suppress RANK Ligand

Bisphosphonates

-inhibit Bone resorption
inhibit with Farnesyl synthase to prevent ruffled border formation
-this reduces osteoclast ability

Anti RANK Ligand

-or also called RANK Ligand Antibody
-prevents precursor of osteoclast from differentiating

Osteopetrosis

-increased bone mass
-osteoclast mutation prevents bone resorption

Osteoclerosis

-decreased bone mass
-osteoblast & osteocyte mutation so their function is reduced

Bone Cancer

-bone is common site for cancer metastasis
-associated with hypercalcemia, nerve compression, bone pain, and fracture

Treatments for Cancer Can Cause Osteopenia & Osteoporosis

1) all treatments cause hypogonadism
2) elevated bone turnover
