catalase test
Detects the catalase enzyme.
Helps differentiate Gram + cocci.
what does catalase do?
Breaks down H₂O₂ (hydrogen peroxide) into H₂O (water) + O₂ (oxygen gas).
Protects bacteria from toxic Reactive Oxygen Species (ROS) and helps them survive in oxygen-rich environments.
What is a positive catalase test?
bubbles (oxygen is released)
What does the oxidase test detect?
The oxidase enzyme (cytochrome c oxidase).
What is the role of oxidase?
It is the last enzyme in the electron transport chain (ETC) used during aerobic respiration to make ATP.
What is a positive oxidase test?
Purple/dark blue color change.
What does the starch hydrolysis test detect?
The amylase enzyme
What does amylase do?
Breaks down starch (a complex sugar).
What is a positive starch hydrolysis test?
Clear halo around the bacteria (starch was broken down).
halo= no starch, amylase is present
no halo= starch, no amylase
