Do we know for sure(does it say in the book) that the characters Beatrice and Benedick had a romantic relationship in the past?
No, however, we can easily drive that they at least had heard about each other and their personalities. It is likely that they had interactions before the begining of the play, but we do not know if they were romantic.

“I have known when there was no music with him but the drum and the fife(military), and now had he rather hear the tabor and the pipe(love)… and know he is turned orthography(fancy speaker)… I will not be sworn but love may transform me into an oyster, but I’ll take my oath on it. Benedick; Basically, he is saying that if he ever fell in love, he would go from being a soldier to being a man in love which he considers foolish. He does not want love to turn him into someone with no backbone, like a thing as simple as an oyster. After he says all this, he goes on to talk about his perfect woman that is unachievable.
Who says, “Is it not strange that sheep’s guts should hale out of men’s bodies?”, and what does it mean?(pg 67)
Benedick; This is iorny because it is usually sheep's guts that are used for musical instruments strings and it would be even more inhumane if they used human guts.

Because he has told himself that she is dead, so to see her in person again makes his brain confused so he can not believe it.
Why are Beatrice and Benedick always jumping around the word love?
Both of these characters are scared of falling in love and the commitment/trust that come with it. They do not want to become heartbroken.
Why does Don John flee?
becuase he does not want to deal with the consequences of his actions.
Why did Leonatono want to lie about Benedick loving Claudio?
He did not want to disgrace his name.
