Three types of Staining
Simple
Differential
Special
Simple Stains
Aqueous or alcohol solution of a single basic dye
Purpose: Highlight the entire microorganism so that cellular shapes and basic structures are visible
Procedure: Stain is applied to the fixed smear → washed off → dried & examined
Mordant
Chemical added to a simple stain solution to intensify the stain
Coat structures (flagellum) to make it thicker & easier to see after it’s stained with a dye
Ex: CV, carbolfuchsin, methylene blue, safranin

Differential Stains
React differently with each type of bacteria and thus can be used to distinguish them
Ex) Gram-stain & Acid-fast stain
Acid-Fast Stain
Bind ONLY to bacteria with waxy material
