When does replication occur?
Replication occurs during S Phase
How many replicons are present in Eukaryotic Replication?
Several Replicons are activated at specific times during S Phase.
What does ARS stand for? What does it do?
Autonomous Replicating Sequence, a Sequence that covers ability to replicate efficiently
What is the structure of ARS?
It is an AT rich sequence that is 50 base pairs long, it also contains 4 domains which are A, B1, B2, B3
Which domain is conserved in ARS?
Only the A domain is conserved to generate a consensus sequence that is 11bp long
What does ORC stand for?
Origin Recognition Complex
What is the ORC? What does ORC do?
It is a complex of 6 proteins and it contacts domain A and B1 elements throughout the cell cycle. It targets ARS.
What are the Licensing Factors discussed?
Cdc6 and MCM
What are the functions of Cdc6?
It is an unstable protein synthesized only in G1 and binds ORC, it allows for MCM to bind. It is only recruited to Unlicensed Origin
What are the functions of MCM?
It binds to ORC for initiation to occur, it gets inactivated after replication is done.
When are LFs present? When are they inactivated/destroyed?
They are PRESENT BEFORE replication, they are inactivated/destroyed AFTER replication; Another round of replication cannot occur until LF re-enters nucleus after nuclear membrane.
TERM
When is Cdc6 degraded?
DEFINITION
After initiation of replication
TERM
When and Where? is MCM inactivated?
DEFINITION
Inactivated in the nucleus after passage into S-Phase.
