COMPLEMENT SYSTEM

MONOCLONAL ANTIBODY

• Very specific antibody derived from a single antibody producing cell that has been cloned or duplicated

• HYBRIDOMA:

  • cell line resulting from the fusion of myeloma cell and plasma cell

  • Mouse is immunized with a certain antigen

  • Mouse's spleen cells are harvested and combined with myeloma cells in the presence of Polyethylene glycol

  • Cells placed in culture with: Hypoxanthine, Aminopterin, Thymidine

  • Myeloma cells and spleen cells die; only the hybridoma cells will survive

  • Clinical and research application: Diagnosis and treatment of disease

COMPLEMENT SYSTEM

• Majority are synthesized by the liver, except:

  • C1 — epithelial cells of intestine

  • Factor D — adipocytes

• Present in highest concentration in plasma and key component to all pathways — C3

• Heat inactivation: 56C for 30 mins

• After 4 hrs delay, reinactivate at 56C for 10 mins
