how do we streak using the whole plate methods

  1. carry out the primary inoculum, moving swab or wire back and forth (rotating utensil) to cover a decent section of the plate

  2. using a sterile loop, drag the primary inoculum across the plate 5 times (lift up after each line)

  3. repeat for sections 3 and 4

  4. for section 5, begin in section 4 and then drag out and zig zag up through the empty space (not lifitng at all)

explain how to use a graduated pipette

  1. open up the end of the pipette sleeve

  2. holding the handpiece in dominant hand, attach the pipette

  3. fully remove the sleeve from the pipette

  4. place pipette end into the bottle/tube (not touching the sides)

  5. ensure pipette remains vertical

  6. use handpiece to collect correct amount (reading the meniscus)

  7. place the sleeve back on the pipette when done

  8. then remove the pipette from the handpiece

  9. place sleeved pipette end in the yellow general waste

explain how to use a semiautomatic pipette

  1. unlock the pipette and ensure it is set to the correct amount

  2. get out box of clean tips

  3. open box and push the handpiece into a tip (gap to shoulder is normal)

  4. immediately close the tip box and move it aside

  5. push the plunger to stop 1

  6. place into bottle/tube and slowly release

  7. remove

  8. place into next tube/bottle

  9. push to second stop

  10. remove

  11. release

  12. eject tip into sharps bin

what do we need to consider if using yellow pipette

the tips are shorter, so we must not let our sterile pipette handpiece touch the bottle edges

to do this we hold the bottle on an angle to encourage liquid closer to the opening
