What is a water bath?
A piece of equipment that holds heated water with a temperature control interface that is used to heat samples in an even manner.
What are water baths used for?
temperature sensitive samples, flammable chemicals, experiments that have to be at set, regulated temperatures
What water should be used in the water bath?
DI water. Tap water contains minerals and microbes that can cause buildup on equipment and contamination. MilliQ water is aggressive and can corrode the interior of the bath.
What is the minimum water level in a water bath?
1/3 the height of the fill line
When adding water back into the water bath, what else should be included?
a few drops of water bath treatment to prevent microbial growth
What happens if water bath water touches the caps of sterile media?
this can compromise the sterility of the media and the viability of the cells you are growing
What are the two water baths in the croyle lab and where are they located?
The Precision Reciprocal Shaking water bath in 4.110 and the VWR water bath in 4.128
What is the Reciprocal Shaking water bath mainly used for?
conducting dissolution tests and heat inactivating animal serums including FBS
Which water bath has a temperature that can be adjusted?
The reciprocal shaking water bath.
When should you turn on the precision reciprocal shaking water bath?
at least 30 minutes before your experiment
What functions does the reciprocal shaking water bath have?
the ability to steadily shake and mix samples while maintaining a constant temperature
What should you check before using a water bath?
the water levels and cleanliness
how do you set the temperature/rotation speed on the reciprocal shaking water bath?
hit the select/enter button, use the arrow keys to set temperature, and hit the select/enter button again
how do you start or stop the shaking function of the reciprocal shaking water bath?
hitting the start/stop button
What is the VWR water bath used for?
thawing and warming solutions used for cell cultures
Can the temperature of the VWR water bath be adjusted?
NO. must stay at 37*C at all times. This water bath is never turned off.
Where are conical tubes placed in the water bath>
in the appropriately sized conical tube rack
Where are eppendorf tubes placed in the water bath?
in the foam floating racks
Where are bottles placed in the water bath?
directly in the bath, weighted down with the small blue weights next to the water bath.
when removing samples from the reciprocal shaking water bath, what should you do?
turn off the water bath
when removing samples from the VWR water bath, what should you do?
leave the water bath on and cover it with the lid.
What are the steps for cleaning the water baths?
turn them off, allow the water to come to room temp, cart the bath to the nearest sink, remove and wash all pieces, wash the baisin with liquinox, wipe basin with a kim wipe, spray everything down with EtOH, add water back in after dry with water bath treatment.
