What is partitioning?
The distribution of a substance between two immiscible phases
How is the partition coefficient calculated?
conc in the organic phase/conc ion the aqueous phase
How is P calculated?
Drug added to separating funnel containing two immiscible liquids
flash shaken for up to 1 hour then left so two phases separate out
conc in aqueous phase determined via titration/spectroscopy
conc in organic phase determined by subtraction
most commonly used liquids are octagon and h20
What is LogP?
Gives indication of the lipophilicity of a drug
Higher number = hydrophobic
What is Ion pairing?
Ionised drugs pair with opposite ions creating a neutral complex that can permeate membranes
