chemiosmosis
the ability of a cell to use proton notice force to do work. the “work” performed is changing of ADP to ATP through phosphorylation using ATP synthase
chemiosmotic theory
electron transport has 2 roles: transport electrons down the chain to form water, and pump hydrogen ions out to build proton motive force and make ATP by chemiosmosis
came up with chemiosmosis theory
Peter Mitchell
evidence for chemiosmotic theory
pH is higher in the matrix than the inner membrane - supporting idea of H+ being pumped out (high pH means low H+). also noticed some things when mitochondria was exposed to detergent and dinitrophenol respectively
mitochondria exposed to detergent
no ATP was made but water was. detergent destroyed the lipid bilayer and a gradient could not be established
exposed mitochondria to dinitrophenol
made water but no ATP, causes the uncoupling of oxidative phosphorylation, producing heat rather than ATP
