What is a positive test result?
dark purple
What is a negative test result?
light green
Why are some results more pronounced over others?
due to varying concentrations of the antigens/antibodies present
more antibodies = more color = more viral infection
What basic principles of antibody-mediated immunity are utilized in an ELISA assay?
AB will attach to certain antigens it is geared to react/bind to
binding will lead to color change of purple versus green where the positive is the liquid turning green in response to the binding and its label
How does an ELISA indirectly detect infection by a disease-causing agent?
detects infection through the use of secondary AB that target the primary antibodies instead of the antigen or disease-causing agent
What is the function of the secondary antibody and chromogen in an ELISA?
secondary AB → used to bind to the primary AB that binds to the simulated antigen
secondary AB has a label/enzyme that will react to the chromogen and allow for a change in color
How to perform ELISA assay?
3 wells for 3 repeat trials of each sample (controls and patient samples)
3 drops of each sample → 3 drops of secondary antibody → 3 drops of simulated chromogen
How long after adding chromogen will color change?
5-10 minutes at room temperature
