What does it measure?
Absorbance (how much light passes through a solution)
Wavelength
Color of light — set to lambda max (peak absorbance for your pigment)
Lambda max
The wavelength where your sample absorbs the MOST light
Calibration (blank)
Use water or buffer — this is your "zero" (0 absorbance)
Cuvette
Small glass/plastic tube that holds the sample
Absorbance vs concentration
Higher concentration = higher absorbance (direct relationship)
Standard curve
Known concentrations → plot absorbance → find unknowns
