Inoculate the broth in 0.1 ml of water, 1 mL, and 10 ml, then incubate at 37 degrees Celsius, and wait 48 hrs

A positive test has gas and growth in the test tube

Take water from the positive presumptive test and spread on emb plate at 37 degrees. And wait 24 hours

Emb test for gram negative bacteria (growth will occur ) and lactose fermenting bacteria (metallic green color)

Take sample from EMB and streak on phenol red lactose agar plate. If it turns yellow then it is acidic and lactose fermenting. Then take a sample from the plate and do gram staining and spot staining. The organism should be gram _ and non spore forming
