What does DNA pol. do?

What are some sources of DNA damage?
chemicals, UV light, oxygen free radicals (by producti of metabolism)
Why is damage to DNA bad?
if damaged DNA is not repaired correctly, the damaged DNA will not pair correctly during replication
damaged DNA can eventually lead to mutations
Mutation
a stable change in the nucleotide sequence of a genome
mutations are not the same as damage, there is no way to fix mutations
What is an example of a DNA repair mechanism?
Nucleotide excision repair
UvrABC complex recognizes damage, cuts out damaged area of DNA (large chunk)
DNA pol. 1 fills in gap, DNA ligase seals gap
