takes 6-12 weeks for clinical effect, will not see an immediate effect

occurs more with use of PTU than MTX, which is why MTZ is USUALLY preferred to PTU

during the 1st trimester of pregnancy -> less PTU crosses the placenta than MTZ and PTU can be supplied with levothyroxine to ensure fetus has enough TH to develop
