Amphotericin B
MOA: Polyene antibiotic; binds fungal cell membrane and increases membrane permeability. Use: Systemic and subcutaneous fungal infections (PO and IV). SE: "Ampho-terrible" — renal and multi-organ toxicities.
Nystatin
MOA: Polyene antibiotic; binds fungal cell membrane and increases membrane permeability. Use: Candida infections. SE: Oral or topical use only.
Clotrimazole
MOA: Inhibits ergosterol synthesis. Use: Candida infections of mouth, throat, vagina, vulva; dermatophyte infections. SE: Oral and topical formulations.
Fluconazole
MOA: Inhibits ergosterol synthesis. Use: Cryptococcal meningitis (AIDS patients), vaginal candidiasis. SE: Excellent CSF penetration.
Ketoconazole
MOA: Inhibits ergosterol synthesis. Use: Seborrheic dermatitis. SE: Oral and topical formulations.
Terbinafine
MOA: Inhibits ergosterol synthesis. Use: Onychomycosis. SE: Typically used orally when topical therapy has failed.
