Experiment 9 Incubation Parameters
The standard environment for growing cultures in this experiment is 37∘C for 18−24h.
Lactase
An enzyme of interest in Part A (Carbohydrate digestion) used by E. coli to break down lactose.
Sucrase
An enzyme of interest in Part A (Carbohydrate digestion) used to break down sucrose into its products.
Amylase
The enzyme of interest in Part B responsible for starch digestion on a Starch Agar plate.
DNAase
The enzyme of interest in Part C that breaks down DNA, tested using a DNA plate with organisms such as PA, Ca, and Sm.
TSA Slant
Tryptic Soy Agar slant used in Part D to test for Catalase activity in organisms like E. coli and B.m.
SIM Tubes
Specialized medium used in Part E to test for Sulfur-indole-motility (SIM) using an inoculating needle.
Cysteine desulfurase
One of the enzymes of interest in Part E (SIM test) involved in sulfur reduction.
Tryptophanase
An enzyme of interest in Part E (SIM test) involved in indole production from the breakdown of tryptophan.
Urease
The enzyme of interest in Part F, tested by gliding an inoculating loop along the agar surface to observe breakdown products.
Starch Agar Plate
The specific medium used in Part B to detect starch digestion by amylase-producing organisms.
Glucase
