Gram stain
A procedure used to determine gram reaction, size, and shape using reagents including crystal violet(purple),iodine(remain purple), gram's declorizer(gram pos remain purple & gram neg colorless), and safranin(basic;gram pos remain purple & gram neg pink)
Acid fast stain
Detect Myobacterium. Counterstain with Methylene blue. Acid fast bacteria appears pink & non acid fast bacteria are blue
Shaffer Fulton Endospore stain
Detect endospores in gram positive cells. Malachite green to dye endospore & counterstained with safranin(cells turn pink)
Mannitol Salt agar (MSA)
A medium used for Staphylococcus isolation that tests for salt tolerance (7.5%NaCl) and mannitol fermentation; positive results turn yellow due to low pH and negative results remain red.
Bile esculin
A test used to detect members of Genus Enterococcus utilizing Esculin and ferrous salts; a positive result is a Black precipitate, while a negative result remains amber.
Catalase
A test to determine if an organism contains the enzyme catalase using 3% hydrogen peroxide; a positive result is the release of oxygen bubbles (O2).
Coagulase
A test used to differentiate species of Staphylococcus (specifically S. aureus) by checking for the formation of a clot in plasma; the tube method is more reliable than the slide method.
Blood Agar
A medium used to observe hemolysis: Alpha (green color around colonies), Beta (complete clearing), and Gamma (no hemolysis).
CAMP reaction
A test that identifies a zone of enhanced hemolysis shaped like a triangle between the organism and Staphylococcus aureus.
Novobiocin sensitivity
A test used to differentiate staphylococci species, specifically distinguishing sensitive S. epidermidis from S. Saprophytius.
Bacitracin sensitivity
A test used to differentiate and ID beta-hemolytic staphylococcus Indicators Group A staphylococcus &Streptococcus pyogenes
Lancefeild A Immunochromatography test
Determine presence of Group A streptococcal carbohydrate antigen Indicators: streptococcus & pyogenes
TSA (Tryptic Soy Agar)
A medium used for quadrant streaks using sterility testing to achieve isolation of pure cultures in the 3rd or 4th quadrant and to check for pigmentation of colonies.
Eosin methylene blue agar (EMB)
A selective and differential medium that isolates and distinguishes bacteria. Selective dye Eosin Y & Methylene blue
SIM (Sulfide-Indole-Motility)
detects sulfur/H2S (blackening of medium), indole/tryptophasnase (red color in added reagent), and motility (fuzzy appearance where stab is not visible). Kovac reagent
Motility agar
A medium with Tetrazolium salt used to see if bacteria move independently via flagella.
