TDT - tube method
Inoculated menstruum is distributed in small-diameter test tubes, heated in an oil bath, then plunged into water at below 70F, then after cooling the contents are plated and the number of survivors is determined.
TDT - can method
cans are heated in small, accurately controlled steam retorts
Tank method
inoculated substrate is placed in the heating chamber
is agitated to desired temperature
Flask method
this method is for studying resistance of bacteria at temperatures below the boiling point of water
a three-neck flask (Woulff bottle) is employed as the substrate container
Thermoresistometer method
was designed by Stumbo (1948) particularly for studying resistance of bacterial spores to temperature in the higher range (above 240F).
applicable for studying resistance temps over the range of 215 to 300F
