Ammonium diphosphate - Alkaline
Simmons citrate agar

Citrate Interpretation
Positive test = Blue
Alkaline when pull ammonium off as carbon source
Nitrate Test Add Ins
Nitrate A - Sulfanilic acid
Nitrate B - Alpha-naphthylamine
Zinc
Sulfanilic acid
Converts p-diazonium benzene to sulfonic acid
Alpha-naphthylamine
Turns sulfonic acid into red pigment
Zinc
If nitrate present, it reduces it to nitrite

Nitrate Interpretation
Red after Nitrite A and B = Positive for Reduction
Red after Zinc = Negative
Colorless after Zinc = Positive for denitrification
Indole Test Looks for
Tryptophanase enzyme
Breaks down tryptophan into ammonia, pyruvic acid, and indole
Tryptic soy agar broth

Red = positive
Yellow = negative
