What does SDS do?
It denatures proteins into a linear structure and applies a uniform negative charge, proportional to size.
SDS
sodium dodecyl sulfate
2-D gel electrophoresis differentiates based on [ ] and then [ ]
charge, size
2-D gel electrophoresis sorts for charge by
having a pH gradient. proteins will migrate only as far as their isoelectric point.
agarose has [ ] pores and is used for [ ] electrophoresis
larger, DNA
polyacrylamide has [ ] pores and is used for [ ] electrophoresis
smaller, protein
purification based on solubility
centrifugation, salting out
purification based on size
gel permeation chromatography, SDS-PAGE
purification based on charge
ion exchange chromatography, gel electrophoresis, isoelectric focusing
purification based on binding specificity
affinity chromatography
