Cellular Location
Cytoplasm of the cell
Primary Organs
Liver (the main factory), Adipose tissue (the storage warehouse), and Lactating Mammary Glands
Hormonal Control
Heavily stimulated by Insulin (the fed-state hormone). Inhibited by Glucagon and Epinephrine
The Building Blocks
The pathway requires Acetyl-CoA (the carbon source) and NADPH (the reducing power, primarily from the HMP Shunt)
The Step-by-Step Pathway
Step A: The Citrate Shuttle (Escaping the Mitochondria)
Step B: The Rate-Limiting Step (Activation)
Step C: The Assembly Line (Elongation)
Step A: The Citrate Shuttle (Escaping the Mitochondria)
Fatty acid synthesis happens in the cytoplasm, but the raw material (Acetyl-CoA) is produced inside the mitochondria. Acetyl-CoA is physically incapable of crossing the inner mitochondrial membrane.
The Disguise: Inside the mitochondria, Acetyl-CoA binds with Oxaloacetate (OAA) to form Citrate.
The Escape: Citrate easily crosses the mitochondrial membrane into the cytoplasm.
The Unpacking: Once in the cytoplasm, the enzyme ATP Citrate Lyase cleaves Citrate back into Acetyl-CoA and OAA. The Acetyl-CoA is now ready for lipogenesis.
Step B: The Rate-Limiting Step (Activation)
Before Acetyl-CoA can be assembled into a long fat chain, it must be activated into a 3-carbon molecule called Malonyl-CoA.
The Enzyme: Acetyl-CoA Carboxylase (ACC). This is the single most important, heavily tested enzyme in this pathway!
The Cofactors Required: ACC requires three things to work.
Cofactors required in step B
Board Mnemonic: "ABC"
ATP (Energy)
Biotin (Vitamin B7)
Carbon Dioxide (CO2)
Step C: The Assembly Line (Elongation)
Now that we have Malonyl-CoA, we can start building the fat chain.
The Enzyme: Fatty Acid Synthase (FAS). This is a massive, multi-functional enzyme complex.
The Process
The Energy Cost
The End Product
The Process
FAS takes an initial Acetyl-CoA molecule and repeatedly adds 2-carbon units from Malonyl-CoA.
The Energy Cost
Every time FAS adds 2 carbons to the growing chain, it consumes 2 molecules of NADPH.
The End Product
The process stops automatically when the chain reaches 16 carbons in length. This final product is Palmitate (Palmitic Acid), a 16-carbon fully saturated fatty acid.
Triglyceride Synthesis (Packaging for Storage)
Palmitate cannot just float around freely; it is toxic to cells in high amounts. It must be esterified (packaged) onto a glycerol backbone to form a Triglyceride (Triacylglycerol).
The Backbone in triglyceride synthesis
The cell uses Glycerol-3-Phosphate
Exam Pearl: Where does this come from? In the liver and adipose tissue, it is a direct offshoot of glycolysis (from DHAP). This perfectly links carbohydrate metabolism to fat storage: You cannot store fat without running glycolysis!
The delivery in triglyceride synthesis
The liver packages these newly minted triglycerides into VLDL (Very Low-Density Lipoprotein) particles and ships them through the blood to adipose tissue for permanent storage.
Regulation: The "On/Off" Switches
Examiners love testing how the body prevents Lipogenesis (making fat) and Beta-Oxidation (burning fat) from happening at the exact same time. This is controlled by covalent and allosteric regulation.
Covalent Modification (Hormones): Insulin (High Energy), Glucagon/Epinephrine (Low Energy/Stress)
Allosteric Modification (Metabolites): Citrate, Palmitoyl-CoA
Insulin (High Energy)
Triggers a phosphatase enzyme to strip a phosphate off Acetyl-CoA Carboxylase (ACC), turning it ON
Glucagon/Epinephrine (Low Energy/Stress)
Triggers a kinase enzyme to phosphorylate ACC, turning it OFF.
Citrate
Acts as a powerful feed-forward activator. If citrate levels are high in the cytoplasm, it means the Krebs cycle is backed up with too much energy, telling ACC to speed up and store it as fat.
Palmitoyl-CoA
The end product of the pathway. It acts as a negative feedback inhibitor, telling ACC to slow down because enough fat has been made.
The Ultimate Gatekeeper: Malonyl-CoA
This is a massive, high-yield board concept. When ACC creates ‘blank’, this molecule does not just build fat; it acts as a lock on the mitochondrial door.
‘blank’ powerfully inhibits CPT-1 (Carnitine Palmitoyltransferase I).
Because CPT-1 is blocked, existing fatty acids cannot enter the mitochondria to be burned via Beta-Oxidation. This guarantees that when you are building fat, you cannot burn fat.
Non-Alcoholic Fatty Liver Disease (NAFLD)
In states of chronic caloric excess and hyperinsulinemia (Type 2 Diabetes / Insulin Resistance), Lipogenesis is permanently stuck in the "ON" position. The liver synthesizes triglycerides faster than it can package and export them as VLDL. Fat droplets accumulate inside the hepatocytes, leading to severe liver inflammation (steatohepatitis) and eventually cirrhosis.
Biotin (Vitamin B7) Deficiency
Because Biotin is a mandatory cofactor for Acetyl-CoA Carboxylase, severe biotin deficiency shuts down lipogenesis. This is rare but can be triggered by massive consumption of raw egg whites (which contain Avidin, a protein that irreversibly binds and traps Biotin in the gut).
