Normal White Blood Cell Count
Feline: 3.8-19 10³ uL
Canine: 5.9-16.6 10³ uL
Total White Blood Cell formula
Average x 50/ 1000 = __________ 10³ uL
Relative count of WBC differential
Ex: 80 neutrophils = 80% for the relative count
Absolute count of WBC differential
Relative % / 100 x WBC count (10³ uL) = ________ x 10³ uL
Stains for WBC
Known as Diff Quik in 4 components:
1.) Fixative (aqua blue) - 95% methanol, fixes cells to slide
2.) Eosin (reddish orange) - Acidic pH, attracted to alkaline cell cytoplasm. Stains hemoglobin and eosinophilic granules pink to reddish
3.) Methylene Blue or New Methylene Blue (purple blue) - Alkaline pH, attracted to acidic (DNA, RNA). Stains nuclei and basophilic granules blue to purple
4.) Rinse - Distilled water to rinse stain off slides
Semi-quantification scale
Scale of 1+, 2+, 3+, 4+
Indicates the relative percentage of cells with changes
◦ 1+ = 5% to 10% of cells
◦ 2+ = 10% to 25% of cells
◦ 3+ = approximately 50% of cells
◦ 4+ = more than 75% of cells
