gel electrophoresis

rflp process:

  1. extraction
  • obtain dna
  1. restriction
  • cut dna into fragments by restriction enzymes
  1. separation
  • gel electrophoresis

electrophoresis: a method of separating molecules by pacing them in a gel through an electric field

  • used to separate DNA, RNA, and proteins

dna is negatively charged

  • attracted to positive end of gel

separation is based on size of fragments

  • smallest travel furthest
  • largest travel least

buffer

add buffer solution because it conducts electricity

micropipette:

  • holds exactly 40 microliters
  • never hold upside down

fill micropipette:

  1. depress
  2. hold
  3. insert
  4. release
  5. remove

fill gel

  1. insert
  2. depress
  3. hold
  4. remove
  5. release
