So what does that mean for the organism? Good. No lactose fermentation. 
When we do not see a color change, you do not look for a bubble litter, too. Okay? Even if there is a bubble in the derm tube, it's not from lactose fermentation, right?

We are going to swab our hand, swab it pretty good. Okay, you can get around the nails, under the nails, anything like that. You're then going to take that slob and do a simple streak in the before. throw that swab in the biohazard bin. 
You gonna either wash your hands, like you normally would. Don't do extra, right? This is really to see how good you are doing for your typical. 
So if you usually watch games in 2 seconds, then do that today. Don't be standing back there, singing happy birthday twice. You know, you don't do that at sync. 
And then for those of you that are using the hand sanitizer, hey, you're gonna stop your hand before, use hand sanitizer, and then swab your hand, pick it. If you want to use the other hand, you can, right? We are technically removing the microorganisms from one hand as we do that with the swamp, but it is up to you. 
So again, swamp, simple street, and then slob after... with another simple street. Okay? In addition, you are going to have another plate, and on that plate, you are going to be making a long street of staphylococcus arius with a swab, okay? 
Because we're going from a broth. Hey, so make that long street really coat the plate. After that, we are going to divide that plate into four quadrants, and we are going to add four different paper disks with different antiseptics. 
A, one of those antiseptics is peroxide. One of them is 10% bleach. One of them is ethanol. 
And one of them is going to be mouthwashed. We have these sterile paper disks back here. Okay? 
I will have some placed into the mouthwash for you, so you're gonna come back with forceps and grab them out of the mouthwash. The mouth wash will be on a paper towel. You don't want to take a dripping, wet, little paper disk and put it on plate, okay? 
Because it's gonna spread all out and make a mess and ruin our smear. So, you can pull it from the mouthwash, dab it on a paper towel, and then place it on your plate, or the mouthwash. Now, for the other three, each of you can take a bottle of peroxide to your table. 
On your table, you also already have ethanol. On your table, you also already have 10% bleach. So you're just gonna get a paper towel. 
Okay, you're gonna lay your paper discs out. One of them, you're gonna put a couple drops hydrogen peroxide on. The other one, you're gonna squirt a little bit of ethanol on. 
The other one, you're gonna put some 10% each on. Again, same thing. Make sure they're not dripping. 
When you're transferring them to the plate, and you're gonna put each of those three in a quadrant. Once your first and second plate are done, they need to be para filmed, and your whole tables will get stacked into the incubator. You are then free to come over here to the back table, find your unknown. 
Hey, they were in the refrigerators. They all should be healthy, and you are going to inoculate one test today. One test. 
So, if you have gram positive coxy... The first thing that you are going to do is the catalase test. Okay? 
Now again. I can help tell you that you're on the right track, Frank guides you, but if you don't know how to do the Cadillacs test, you can't remember, or you can't remember the purpose that is on you to figure that out from your notes or in the lab manual, because we have already done it. Now, the Catalese test is a rapid test, so today, you are going to see if it is positive. 
Or negative. You guys get an extra step today, because you need to know that rapid test result before you can actually inoculate any sort of media. If your organism is positive, you are going to find the table in exercise 53. 
That table and exercise 53 is going to give you a variety of tests, and it is up to you to choose which one. It doesn't matter what one, you can pick one to eliminate sap Oreus, you can pick one. you think you have something else, right? If it is negative, similarly, you are going to find a table and exercise 54 in your lab manual. 
That table and exercise to be for also has a variety of texts. You choose which test you want to do. Most students for that one start with analysis. 
But again, it is totally up to you, it's a bigger fan out. And then you're gonna inoculate one test, and that's it. Only one test per day, you guys get to do two, because the cattle base is rapid. 
If you have gram positive rods, I put a flow chart on canvas. You guys are extra special. So, if you go to the lab or lecture campus, you will find a folder that says unknown guidelines, and very on positive flow charts. 
You will pull that blow chart up, and we will follow it. Okay, it has the possible species that you can have in a couple of different tests that you could do, but you start at the top, inoculate that media. This one is a motility to start. 
Next week, you'll get those results, and then it will take you either to go to the left or right of that closure. Okay? So the gram positive fraud, and the gram negative rods are a bit more structured. 
Whereas the grandpa's in coxy, we've got a little bit more freedom, okay? So you kind of get to choose really what you are doing or what you're not doing. RAM negative rods. 
You guys are going to be going to exercise 55? Or gram negative rocks, it is not a table, like the grandpa's in Foxy. It is a flowchart, okay?
